He instead offered to appear in a public session to discuss the claims against his father, telling reporters: “Let me state as clearly as I can, ‘My father was not involved in my business’.”
His refusal to appear for a private deposition means he now faces charges of contempt of Congress.
Jim Jordan and James Comer, the Republican congressmen leading the House committee investigations into the president, said they would now seek charges against him.
“Hunter Biden today defied lawful subpoenas and we will now initiate contempt of Congress proceedings,” they said. “We will not provide special treatment because his last name is Biden.”
House Republicans have alleged that the president had knowledge of, and benefitted from, illegitimate business dealings by his son.
Republicans running the committee investigations have previously been accused by the White House of a politically-motivated attack on both the president and his son, in an apparent attempt to distract from legal proceedings against Donald Trump.
The committees investigating the president have so far produced no evidence that he benefited from his son’s business dealings. The president himself has described the allegations as a “bunch of lies”.
In his statement on Capitol Hill, Mr Biden Jr said: “For six years, MAGA Republicans, including members of the House committees who are in a closed-door session right now, have impugned my character, invaded my privacy, attacked my wife, my children, my family, and my friends.
“They ridiculed my struggle with addiction, they belittled my recovery, and they have tried to dehumanise me – all to embarrass and damage my father, who has devoted his entire public life to service.
“For six years, I have been the target of the unrelenting Trump attack machine, shouting: ‘Where’s Hunter?’ Well, here’s my answer: ‘I am here’.”
Threatened with contempt
Mr Biden Jr has previously been threatened with contempt proceedings if he did not appear before the committees investigating his father.
His attorney, Abbe Lowell, said that he would not appear for a deposition behind closed doors because Republican representatives could selectively brief his testimony to the press in an attempt to discredit him.
“He is making this choice because the Committee has demonstrated time and time again it uses closed-door sessions to manipulate, even distort, the facts and misinform the American public a hearing would ensure transparency and truth in these proceedings,” he wrote.
Potential witnesses can be held in contempt of Congress if they do not comply with lawful subpoenas.
Separately, it was reported on Sunday that the president feels guilty about the federal prosecution against his son, and believes it would not have taken place if he was not the president’s son.
Mr Biden Jr has been charged with four felonies and six misdemeanours, relating to an allegedly falsified tax return and possession of a gun while using illegal drugs.
